Effects of ATF3 on Berberine-inhibited adipogenesis and obesity

Abstract

    Obesity, as one of the most epidemic diseases, is known as a strong risk factor for T2DM. Berberine (BBR) is well known to have beneficial effects in the treatment of diabetes and obesity. Here, we have investigated the role of ATF3 and its downstream target regulator during BBR-inhibited adipocyte differentiation. BBR reduced body weight, plasma triglyceride, and plasma glucose or insulin levels and improved insulin action in high fat diet-fed rats. In BBR-treated adipose tissues, ATF3 was significantly increased along with the Pref-1 and its downstream target Sox9 that is highly expressed in preadipocyte. Concomitantly, BBR treatment resulted in decreased adipocyte differentiation and delayed the downregulation of pref-1 and sox-9 expression, which subsequently, leads to the reduction of adipogenic gene expression. ATF3-inhibited adipocyte differentiation was accomplished by increasing the expression of Pref-1 and Sox-9. We found that BBR-induced ATF3 may play as an upstream inhibitory regulator for the downregulation of Pref-1 and Sox-9, early markers of adipocyte differentiation, thus leads to the reduction of adipocyte differentiation and obesity.
