Yersinia pestis impairs maturation and activation of dendritic cells in mouse

Abstract

    Purpose: Yersinia pestis, an etiological agent of plague, causes a fatal infectious disease to humans via flea vector. In this study we investigated the effects of Y. pestis grown at the temperature of flea vector (27˚C) or mammalian host (37˚C) on dendritic cells (DCs) maturation and activation.
    Methods: Mouse bone-marrow-derived DCs (BMDCs) were exposed to killed or live Y. pestis grown at 27˚C or 37˚C, respectively. Expression of DC maturation markers along with secretion of cytokines and chemokines was measured. In addition, killed or live Y. pestis?stimulated DCs were co-cultured with splenocytes and production of T helper (Th)1 and Th17 cytokine was also measured.
    Results: Killed Y. pestis cultured at 37˚C reduced DCs maturation markers including CD80, CD 86, and MHC II and inflammatory mediators production including TNF-α, IL-6, MIP-1α, MCP-1, and IP-10 than did Y. pestis cultured at 27˚C. No difference was found in the expression of maturation markers of DCs upon stimulated with live Y. pestis grown at 27˚C or 37˚C. However, secretion of inflammatory mediators was reduced in response to live Y. pestis grown at 37˚C compared with live Y. pestis grown at 27˚C. Production of IFN-γ and IL-17 induced by killed and live Y. pestis grown at 37˚C was much lower than did Y. pestis cultured at 27˚C.
    Conclusions: These results showed that Y. pestis grown 27˚C or 37˚C could induce the maturation and activation of DCs by different mechanism(s).
