Molecular Typing of Mycobacterium abscessus complex by Variable Number Tandem Repeat assay (VNTR)

Abstract

    M. abscessus complex is major Nontuberculous mycobacterium (NTM) causing pulmonary infection. This complex is composed of 3 related species, M. abscessus, M. massiliense, and M. bolletii. Molecular typing is useful for the epidemiological investigation and characterization of NTM, and VNTR has advantage in time and cost. In this study, we analyzed 54 M. abscessus, 46 M. massiliense, 2 M. bolletii clinical isolates, and 2 reference strains by VNTR assay using 7 loci. 104 isolates were classified into 86 VNTR types. The Diversity index of 7 loci was 0.978. So it has strong discriminative power. 86 patterns were grouped into 5 clusters. In cluster 4 and 5, only M. abscessus was included, but in other, two or three species were included. All the M. massiliense hsp65 genotype II isolates was included in cluster 3, but in this cluster, other genotype and species were also included. In our previous study using Pulsed-field gel electrophoresis, hsp65 genotype II-1 was composed separate cluster. In conclusion, though VNTR assay has high discriminative power, it has limit for the classification of M. abscessus complex in the species or intra-species level. (Supported by grants from KCDC)
