Abstract
KVAC103 is a highly attenuated vaccinia virus strain, developed for a third-generation smallpox vaccine at the Korean National Institute of Health (KNIH) by multiple propagations of the Lister strain in Vero cells and plaque isolation. It has a 20-kb deletion in the left wing of the genome and can replicate only in restricted mammalian cell lines. In this study, I tried to improve the replication of the virus and to develop an efficient recombinant virus production method. DNA fragments of the deleted region and the green fluorescent protein (GFP) marker gene were inserted into the thymidine kinase (tk) locus of KVAC103, and replicating mutants were screened. Of the deleted genes, C7L and K1L rescued GFP-positive recombinant virus efficiently. Using a plasmid shuttle vector containing these genes, recombinant viruses could be easily obtained. The recombinant virus replicated efficiently in diverse mammalian cell lines and expressed 10-fold higher levels of GFP antigens. Such a recombinant vaccinia virus production system facilitates the development of recombinant vaccines.
