Abstract
Recent advances in nano-materials and its application sophisticate the sensitivity of conventional analytis methods used in biological fields. Previously, we developed the oligonucleotide-linked immunosorbent assay with gold nanoparticle (GNP), named as GNP-OLISA, which are modified detection method derived from enzyme-linked immunosorbent assay (ELISA) and we demonstrated that GNP-OLISA is 100 fold higher than ELISA in detection sensitivity of cancer biomarker models. In this study, we further modified experimental condition and applied GNP-OLISA to detect Francisella tularensis, the bio-terror agent responsible for tularemia, because current detection methods such as ELISA and real-time polymerase chain reaction for F. tularensis are not sufficient to identify the minimum infectious dose (about 10 colony-forming units (CFU)) of F. tularensis. Using GNP-OLISA, we could detect below 10 CFU of F. tularensis from PBS-diluted sample and limit of detection (LOD) value was 23.47 CFU mL-1. In addition, we can also detected F. tularensis from rabbit-serum spiked sample in the presence or absence of other high-risk pathogen (Bacillus anthracis) and the LOD were 30.79 CFU mL-1 and 29.63 CFU mL-1, respectively. It implies that GNP-OLISA can sensitively identify the bacteria from serum sample as well as crude sample without significant elevation of background signal.
