LPS Prevents Mouse from Infection of Francisella tularensis LVS

Abstract

    Francisella tularensis is high-risk pathogen, which can cause of tularemia. As only about 10 bacteria can lead to disease and early symptoms of tularemia do not reveal their identity, development of vaccine for Francisella tularensis is critical for tularemia prevention. However, there is no official vaccine approved by FDA yet. Recently, LPS purified from Francisella tularensis have good efficacy for tularemia prevention, however detailed in vivo process of prevention by LPS is not completely elucidated yet. For rigorous analysis of Francisella tularensis pathogenesis in LPS vaccinated mice, we visualized the bacteria and analyzed of bacterial burden in infected organs. For visualization of pathogen, we construct dual-labeled bacteria which express both fluorescent and luminescent signals, and analyze each organ infected with Francisella tularensis using molecular imaging method. By intraperitoneal infection of Francisella tularensis, liver, lung and spleen were infiltrated with replicated bacteria. However the replication of bacteria in each organ was drastically suppressed by LPS pre-treatment with intraperitoneal route. Our study suggests that LPS can act as a good vaccine for Francisella tularensis LVS in mouse, and our newly developed imaging method for efficacy test could supply detail of bacterial distribution during vaccination.
