Development of Reference Strains for Bactericidal Efficacy Tests of Commercial Disinfectants/Sanitizers

Abstract

    Strains originated from American type Culture Collection (ATCC) are currently used as reference strains in general test methos. Korean food Additives Codex presents standardization test methods for assessing the effect of commercial sanitizers/disinfectant in Korean which is composed of 3 test methods such an in vitro suspension test, a surface test, and spore suspension test. The methods adapt Escherichia coli ATCC11229, staphylococcus aureus ATCC6538 and Bacillus subtilis ATCC6633 as reference strains. Therefore, the aim of this study is to develop reference strains with Korean cliical isolates for replacement of ATCC strains. From more than 500 strains registered in National Culture Collection for Pathogens (NCCP), 18 candidate strains were selected by bactericidal effect, biochemical test and 16s tRNA sequence analysis. Their sensitivity was tested to six representative sanitizer/disinfectants including ethanol, n-Alkyl(C12-C18)benzyldimethly Chloride, sodium hypochlorite, peroxyacetic acid, iodine and citric acid and the the results were compared with ones of the ATCC references strains. The strains showing same sensitivities with a reference at 100%, 50% and 25% of their maximum permissible concentrations based on Korean food additives codex was finally selected as the alternative to ATCC reference strain, which was confirmed by observing their similar morphologic changes on exposure to sanitizers/disinfectants under electronic microscope. thus, we suggest than E.coli NCCP14134, S. aureus NCCP14339 and B.subtilis NCCp10827 could be applied as the reference strains for authorized test methods. These trials also have significant meaning in therms of development and practical use of domestic bioresources
