Recapitulation of MAGIC consrotium meta-analysis results for HOMA-%B and HOMA-IR in two Korean populations

Abstract
Homeostasis model assessment (HOMA) has been widely used to yield the beta-cell function (HOMA-B) and insulin resistance (HOMA-IR) from fasting insulin and glucose concentrations. In this study, we examined the associations of SNPs with marginal p-value < 0.05 in the MAGIC study for HOMA-%B or -IR by using Korean community-based cohorts (Ansung and Ansan) which were known as KARE. The MAGIC consortium released the summary data for 2.5 million SNPs and, among the 2.5 million SNPs, 221,936 SNPs showed the marginal association (p < 0.05). 27,818 SNPs and 112,123 SNPs were presented in the KARE genotyped SNPs and imputed SNPs. The replicated SNPs were decided by the same direction of beta value in all three study groups (Ansung, Ansan, and MAGIC), the p-value was lower than 0.05 in both Korean cohort (Ansung and Ansan), and in both HOMA models (HOMA1 and HOMA2). 75 SNPs satisfied our criteria at the association with HOMA1-%B and HOMA2-%B, and 36 SNPs satisfied with HOMA1-IR and HOMA2-IR. 4 genes (DGKB, GCK, GLIS3, and FADS1) were highly significant in this study. Interestingly, the GLIS3 was one of the novel loci identified by the MAGIC, and was repeatedly replicated in Han-Chinese and South Asian. In this study, we discovered two novel loci (CTH and KLHL31) which were associated with both HOMA-%Bs and IRs and literature search indicates that the two genes related to insulin secretion or insulin sensitivity. Our finding may help our understanding of glucose homeostasis in humans and may reveal new pathways for diabetes therapeutics.
